AI Urine Sediment Analysis slide preparation

Shannon Mitchell Updated by Shannon Mitchell

Centrifuge the sample

  1. Determine if dilution is necessary (refer to the dilution guide). For more information, view AI Urine Sediment Analysis dilution procedure.
  2. Mix the sample well.
  3. Transfer 1, 2, or 3mLs to the centrifugation tube.
  4. Add the tube to a centrifuge, using a corresponding tube for balance, and spin the sample for two minutes at 450–500RCF. Allow the centrifuge to come to a complete stop before removing the sample.

Prepare the sample

  1. Attach the XactUrine® pipette tip to the micropipette.
  2. Place the pipette with the tip attached in the tube.
  3. Tilt the tube to pour off the supernatant until no liquid is visualized above the stopper.
  4. Return the tube to vertical.
  5. Press the plunger five times or until the pellet is resuspended.

Prepare the slide

  1. Place a pre-printed slide on the template.
  2. Orient the fiducial circle toward the bottom of the template.
  3. Label the slide with patient information on the frosted edge.
  4. Once the pellet is resuspended, place 20µL in the fiducial circle.
  5. Cover with a 22 x 22mm coverslip.
